Neuroprotective Effects of San-Jia-Fu-Mai Decoction on MPTP-induced Acute Model of Parkinson's Disease

摘要Parkinson’s disease (PD) is a debilitating chronic and progressive neurodegenerative disease. Major clinical symptoms of PD include bradykinesia, muscle rigidity, and resting tremor. Halting or reserving neuron degeneration in PD patients is still difficult. San-Jia-Fu-Mai decoction (SJFMD), a traditional herbal formula consisting of 9 ingredients, has long been used clinically to treat neuropsychiatric disorders. However, scientific evidence of the efficacy of SJFMD in PD treatment is lacking. Thus, in the present study, we aimed to investigate the neuroprotective effect of SJFMD and illuminate its molecular action mechanisms on 1-methyl-4-phenyl-1,2,3,6-tetrahy dropyridine (MPTP)-treated mice. The results revealed that SJFMD improved the locomotor behavior, prevented the degeneration of dopamine neurons induced by the toxin in the substantia nigra pars compacta (SNpc) of mice. SJFMD-mediated neuroprotection is associated with a reduction of the number of glial fibrillary acidic protein-positive reactive astrocytes and a strong inhibition of pro-inflammatory genes such as IL-1β and IL-6, and TNF-α. Moreover, SJFMD blocked the inhibition of the anti-apoptotic gene Bcl-2, and the stimulation of the pro-apoptotic genes Bax and caspase-3 in the SNpc. SJFMD also decreased oxidative damages, overproduction of reactive oxygen species and accumulation of lipid oxidation products in dopamine neurons. Taken together, SJFMD exerts a potent neuroprotective effect against MPTP-induced degeneration of nigrostriatal dopamine neurons in mice, through involving downregulation of neuroinflammatory, oxidative and apoptotic processes.
