In Vivo And In Vitro Study On The Therapeutic Effect Of A Chinese Herbal Formula (Szf) For Atopic Dermatitis

摘要Background and Aims: Atopic dermatitis (AD) is a common skin disorder characterized by erythema, eruption, lichenification and pruritus. Chinese herbal medicines have been used to treat AD for a long time. In this study, we aimed to investigate the therapeutic effect of an empirical herbal formula (SZF) for treatment of AD using in vitro and in vivo models of AD.
Methods: Quality control of SZF was performed using HPLC analysis. In the in vivo study, fifteen female BALB/c mice were randomly assigned into five groups(a) naïve control group (NC); (b) DNCB-treated control group; (c & d) SZF (3.15 or 1.05 g/kg) treated groups; (f) dexamethasone (DXM, 3 mg/kg) treated group. After removal of dorsal hair, 2% DNCB was applied in all groups except for NC group to dorsal skin for 2 days (day 1-2). From day 8 to day 29, 0.2% DNCB was applied every 3 days which repeated for 7 times. SZF and dexamethasone were daily administered by gavage (day 15-29). On day 30, blood and skin samples were harvested for biological analysis. The coat characteristics and in-cage behaviours were recorded throughout the study. In the in vitro study, the antiinflammatory effects of SZF were investigated through evaluating the change of nitric oxide (NO), TNF-α and PGE2 in LPS stimulated RAW264.7 cells.
Results: Baicalin and berberine were of high concentration in SZF. The results of in vivo study showed that DNCB could induce skin thickening, lesions and pruritus, as well as increase the level of IgE in serum, while treatment with SZF (3.15 g/kg) could obviously reverse these changes induced by DNCB in mice. In in vitro study, SZF (400 μg/ml) could significantly inhibit the productions of NO, TNF-α and PGE2 in LPS-stimulated RAW264.7 cells.
Conclusions: Our results demonstrated that SZF could effectively ameliorate DNCB induced AD-like skin lesions in mice and inhibit the productions of NO, TNF-α and PGE2 in LPS stimulated RAW264.7 cells.
