摘要Alzheimer’s disease (AD) is the most common form of dementia in elderly people with huge healthcare and economic burden to the society. Magnolol and honokiol are two ligands isolated from Magnolia officinalis, and can cross the blood-brain barrier. Magnolol and honokiol have been found to improve the memory and learning deficits found in SAMP8 mice. Recent study in our laboratory indicated that honokiol could prevent the learning and memory impairments induced by scopolamine in mice. The aim of this study was to compare the cognitive deficits improving effects of magnolol, honokiol and their combination in Aβ1-42-treated rats. After drugs treatment for 21 days, the spatial learning and memory functions were assessed using Radial Arm Maze test. Our results showed that magnolol, honokiol or their combination could significantly improve the spatial learning and memory functions in Aβ1-42-treated rats. Mechanistically, magnolol, honokiol or their combination markedly decrease the protein levels of Aβ1-40 and Aβ1-42 in the hippocampus of Aβ1-42-treated rats. In addition, magnolol, honokiol or their combination could significantly suppress the Aβ accumulation via enhancement of the α-secretase activity and inhibition of the β-secretase activity (BACE-1). Moreover, magnolol, honokiol or their combination could markedly inhibit the neuro-inflammation induced by Aβ1-42 in the hippocampus of rats and the protein levels of p-GSK-3β (Ser9), while increase the protein level of p-Akt (Ser473) in the hippocampus of Aβ1-42-treated rats. Our results demonstrate that the combination of magnolol and honokiol could ameliorate the learning and memory impairments in Aβ1-42-treated rats, indicating that they are promising natural compounds for further development into anti-AD therapeutics.
