Role of the RhoA/ROCK pathway in regulating the migration of mouse enteric neural crest cells

AbstractThe enteric nervous system is composed of cells derived from vagal and sacral neural crest cells. During embryonic development, vagal neural crest cells migrate from the proximal foregut to the distal hindgut while sacral neural crest cells migrate in an opposite direction from the distal to the proximal hindgut. However, little is known about the molecular mechanism regulating the migration of these two groups of enteric neural crest cells (ENCCs) in mice. In the present study, results from transcriptomic analyses through RNA sequencing and immunofluorescence staining on ENCCs showed that molecules along the RhoA/ROCK pathway including RhoA, ROCK, LIMK and cofilin that are important for cytoskeletal modifications and cell migration were highly expressed. Treatment of ENCCs with ROCK inhibitor Y-27632 inhibited the phosphorylation of these proteins, disturbed the RhoA/ROCK pathway and increased the inter-cellular distance between ENCCs. Overnight treatment of ENCC cultures with glial cell-derived neurotrophic factor (GDNF) also inhibited the phosphorylation of cofilin, which was restored to normal 10 hours after the removal of GDNF, indicating that GDNF might modulate the RhoA/ROCK pathway. Using a Förster resonance energy transfer (FRET)-based biosensor, active RhoA was shown to be expressed at the leading edge of migrating ENCCs in vitro, implicating that RhoA might be important for ENCCs to move towards a particular direction. In conclusion, our results indicated strongly that the RhoA/ROCK pathway is important in regulating the migration of ENCCs.
