Anti-inflammatory activities of Pentaherbs formula in allergic asthmatic mice models

摘要Allergic asthma is a chronic allergic inflammation of the lung, characterized by airway hyperresponsiveness (AHR), bronchial wall remodeling, eosinophil infiltration and elevation of IgE levels. The prevalence of allergic asthma varies around the globe; but children with atopic dermatitis, an allergic inflammatory condition of the skin with a prevalence of 20%, have an increased risk in developing asthma as they grow up. Previous in vitro, in vivo and clinical studies showed that the Pentaherbs formula (PHF) consisting of five traditional Chinese herbal medicines, Lonicerae Flos, Menthae Herba, Phellodendri Cortex, Moutan Cortex and Atractylodis Rhizoma at w/w ratio of 2:1:2:2:2, exhibited therapeutic potential in treating allergic inflammation disease on the skin. In this study, an in vivo murine model with ovalbumin (OVA)-induced allergic asthma was used to elucidate the efficacy of PHF. AHR, bronchial wall remodeling, the infiltration of immune cells in the lung and the release of cytokines in the lung and serum of OVA-induced allergic asthmatic mice were examined. Upon the oral administration of PHF for 14 days, the AHR, smooth muscle hypertrophy, mucus secretion by goblet cells were improved. The number of eosinophils and neutrophils infiltrated into the lung were significantly reduced (all p<0.05) in the PHF-treated allergic asthmatic mice. The Th2-related IL-4, IL-33 and Th1-IFN- levels in the bronchial alveoli lavage (BAL) fluid of the mice were also significantly decreased (all p<0.05) upon 8-day PHF treatment. The 14-day PHF treatment could also moderately suppress the release of IL-31 in the BAL fluid of the mice. These findings suggest that PHF can ameliorate allergic inflammation and may attenuate the activation of eosinophils in OVA-induced allergic asthmatic model. This study also reveals that therapeutic interventions of the atopic dermatitis may potentially prevent the development of subsequent atopic disorders.
