Molecular characterization of a unique ARF protein in plant autophagy

摘要ADP-ribosylation factor (ARF) family proteins, one kind of small guanine-nucleotide-binding (G) proteins, play a central role in the cell membrane trafficking. Located in a wide range of organelles, ARFs are under tightly control via guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). ARF family proteins regulate vesicular traffic and organelle biogenesis by recruiting coat proteins, modulating membrane lipid composition and structure of the actin at membrane surface. In mammalian cells, ARF1 mediates the COPI recruitment to budding transport vesicles between Golgi cisterna, whereas ARF6 activates the PIP5K and phospholipase D (PLD), leading to the large increase in PI(4,5)P2 at the cell periphery in regulation of clathrin-mediated endocytosis. More recently, Arfrp1 was shown to participate in the polarized transport of Vangl2 from the trans-Golgi network (TGN) in mammalian cells. The Arabidopsis genome contains more than 21 ARF proteins but relatively little is known about their functions in plants. Here we aim to identify selective ARFs for their functional characterization in plants using a combination of cellular and molecular approaches. We have first carried out subcellular localization study for the plant ARFs in Arabidopsis protoplasts transiently expressing GFP-tagged ARFs and have identified several ARF proteins of unknown function exhibiting distinct subcellular localization for further characterization. Interestingly, one of the ARF family proteins, termed ARFX in this study, was shown to colocalize with autophagy markers. On-going studies focus on the functional roles of ARFX in plant autophagy using cellular, biochemical, molecular and genetic approaches. Here I will present an update on this study.
