AbstractGerm cells are responsible for generating new lives and tranferring the whole genetic and epigenetic
information to the progeny. For the germ cell generation, a series of sophisticated and elegant developmental
procedures happen, including specification of primordial germ cell (PGC) from epiblast cells (EpiCs), PGC
proliferation and migration to the gonadal ridges and PGC maturation into gametes. It is extremely difficult to
study the molecular mechanism of PGC development in vivo, as only minute quantities of PGCs exist in an
embryo. During the past ten years multiple research groups in the world have put enormous efforts to
reconstruct the germ cells in vitro. Only in recent years, Saitou’s group successfully derived functional primordial
germ cell (PGC) -like cells from pluripotent stem cells and obtained offspring from these cells. The success of
this study suggests an interesting foundation to investigate and reconstitute germline development in vitro. Due
to the limited number of female germ cells, we decided to induce female germ cells from Oct4-GFP
embryonic stem cells. Our preliminary data suggests that Oct4-GFP ES cell is a good model to induce PGC-like
cells in vitro. Female Oct4-GFP PGC-like cells can survive well in the ovary and express typical germ cell
markers. We will further check whether healthy offspring can be obtained from the Oct4-GFP PGC-like cells in
the future.
