Role of Transient Receptor Potential Canonical 5 (TRPC5) Channels in Vascular Tone Regulation

摘要Intracellular Ca2+ concentration in endothelial cells (ECs) and vascular smooth muscle cells (VSMCs) plays an important role in vascular tone control and blood pressure regulation. Previously, most research focused on voltage-dependent Ca2+ channels which mediate Ca2+ influx and vascular contraction. However, recent focus has been shifted to TRP channels. The aim of our study is to investigate the role of TRPC5 channels in vascular tone regulation.
In present study, immunohistochemical staining results demonstrated that TRPC5 is expressed in both ECs and VSMCs which is in consistent with immunoblotting results. Isometric tension of arteries was measured using wire myograph. In carotid arteries (CAs), selective TRPC5 inhibitors clemizole and ML204 attenuated endothelium-dependent contraction (EDC) in a dose-dependent manner. Importantly, EDC in CAs from TRPC5 knockout (KO) mice was substantially lower compared with that of wild type (WT) mice. These results indicate that TRPC5 contributes to EDC response in mouse CAs. Attempts were made to determine which cyclooxygenase (COX) mediates EDC in CAs. In both TRPC5 WT and KO mice, NS-398 and indomethacin abolished EDC but VAS. Ex vivo culture of aortic rings from WT mice, incubation with tunicamycin increased ER stress markers expression, including GRP78, XBP1, ATF6, ATF4 which was inhibited in aortas from KO mice. Incubation with tunicamycin also induced enhanced COX-2 expression which was attenuated in aortas from KO mice.
In conclusion, our studies suggest that TRPC5 contributes to EDC response in mouse CAs. Furthermore, TRPC5 deficiency inhibits ER stress.
