Anti-inflammatory activities of Pentaherbs formula, berberine and gallic acid in atopic dermatitis

AbstractAtopic dermatitis (AD) is an incurable allergic skin disease. We have found that

traditional Chinese medicine, Pentaherbs formula (PHF), shows promising therapeutic

potential in treating AD children with fewer side effects. Herein, we investigated the in

vitro anti-inflammatory activities of the PHF and its potential active ingredients, berberine

and gallic acid, by measuring the cytokine released in the supernatant of the co-culture of

eosinophils and dermal fibroblasts. Berberine could suppress pruritogenic IL-31- and AD-

related alarmin IL-33- induced pro-inflammatory cytokine IL-6 release from eosinophils to

a level comparable to its negative control (p > 0.05). However, the addition of PHF did not

show any significantly suppressive effect on IL-31- and-IL- 33-stimulated eosinophils-

dermal fibroblast co-culture (p > 0.05). Yet, gallic acid could significantly suppress IL-31-

and-IL- 33-induced IL-6 release from co-culture (p < 0.01). Together, our data suggest that

the berberine and gallic acid in the PHF may have a crucial anti-inflammatory role in

treating AD by attenuating the activities of eosinophils interacting with dermal fibroblasts.
