AbstractBackground: Atopic dermatitis (AD) is an incurable allergic skin disease. We have found that a

traditional Chinese medicine, Pentaherbs formula (PHF), shows promising therapeutic potential

in treating AD children with fewer side effects. Herein, we investigated the anti-inflammatory

activities of the PHF and its potential active ingredients, berberine and gallic


Methods: A co-culture system was established to evaluate the interaction between

eosinophils and human dermal fibroblasts. ELISA assays were used to measure the release of

cytokines and chemokines in the supernatant of the co-culture system.

Results: Berberine

could significantly suppress IL-31 and IL-33- induced pro-inflammatory cytokine IL-6 release

from eosinophils alone by half. However, the addition of PHF did not show any significantly

suppressive effect on IL-31- and-IL- 33-stimulated eosinophils-dermal fibroblast co-culture (p >

0.05). Yet, gallic acid could significantly suppress IL-31 and IL-33- induced release of AD-

related inflammatory IL-6 and chemokine CXCL8 from co-culture to a level comparable to the

hydrocortisone treatment group.

Conclusion: Together, our data suggest that the berberine

and gallic acid in the PHF may have a crucial anti-inflammatory role in treating AD by

attenuating the activities of eosinophils interacting with dermal fibroblasts.
