Asymmetric mRNA Expressions of Ladybird Homeobox 1 in Paravertebral Muscles of Chinese Patients with Adolescent Idiopathic Scoliosis

摘要Introduction: Adolescent idiopathic scoliosis (AIS) is the most common paediatric spinal deformity, affecting 3% of
adolescents. Recent genome-wide association studies have demonstrated a strong association between AIS and SNPs near the ladybird homeobox 1 (LBX1) locus. However, the expression of LBX1 in muscle from Chinese AIS patients remains unknown. This study aimed to investigate LBX1 mRNA expression in bilateral paravertebral muscles of Chinese AIS patients and its association with the development of scoliosis.

Methods: Bilateral paravertebral muscles were harvested from 18 AIS patients requiring spinal corrective surgery and
non-AIS controls. Muscle samples were harvested from bilateral paravertebral muscles at the apical vertebral level. The LBX1 mRNA expression was evaluated by the conventional real-time polymerase chain reaction. Besides, LBX1 expressions in bilateral paravertebral muscles were compared in each group. The expression ratio, the expression at the convex side relative to the concave side, was compared between groups.

Results: The expression ratio of LBX1 in paravertebral muscle of the AIS group was higher than the controls. When
the samples were subdivided into convex and concave sides, significant higher LBX1 mRNA expression on the apical
paravertebral muscle of the convex side was observed in the AIS group (p<0.05).

Conclusion: There was a difference on the LBX1 mRNA expression between AIS group and controls. In the AIS group, the expression was asymmetrically observed in the apical paravertebral muscles. This finding suggested the possible functional role of paravertebral muscle in the development or progression of the spinal curvature in AIS.
