Development of Novel Two-Photon Fluorescent Probes for Subcellular Imaging

摘要There has been considerable interest in the development of near-infrared (NIR) absorbing and emitting dyes due to their many promising applications. In particular, these materials are widely used in bioimaging owing to their
advantageous properties that allow reduced auto-fluorescence and deeper light penetration into biological tissues. Two-photon absorption is a third-order nonlinear optical process. TPA molecules can simultaneously absorb
two photons to reach an excited state. Each photon has lower energy with longer wavelength. This allows a deeper light penetration and higher spatial resolution in biomedical imaging. Recently, pyrrolopyrrole aza boron dipyrromethenes (PABDPs) have been prepared by treating diketopyrrolopyrrole with hetero-aromatic amines in the presence of titanium tetrachloride. These compounds exhibit intense absorption and fluorescence in the NIR region, as well as a relatively high photostability. These properties suggest that they are promising candidates for potential applications in molecular optoelectronics and biomedicine. We have recently prepared series of PABDP derivatives that exhibit two-photon absorption properties. With proper modification of these dyes, they can be used for two-photon excited subcellular imaging. In this presentation, we will report the synthesis, one-photon and two-photon spectroscopic properties of these fluorescent probes and their performance in subcellular imaging.
