AbstractMycelial growth and fruiting body development are important topics in mushroom biology and have been analyzed at the molecular level. We have used high throughput methods Differential Display, Expressed sequence tag analysis, and Serial Analysis of Gene Expression to characterize gene expression profiles during development of Shiitake mushroom Lentinula edodes. The kind of genes differentially expressed at the initiation of fruiting body include the following: (1) initiation-stress response and specific signal transduction, (2) reconstruction of proteome-protein degradation, modification and biosynthesis, and (3) switching of biochemical pathways and structural components like hydrophobins. The gene expression profiles during sporulation showed the occurrence of: (1) stress response, (2) biosynthesis of DNA and chromosome, (3) proteome reshuffling (4) production of specific enzymes and proteins. Gene expression profiles revealed by different approaches were compared and were generally consistent.
