Updated classification of viruses in the family Caliciviridae and genus Norovirus

摘要Introduction. The family Caliciviridae includes viruses with a linear positive-sense RNA genome of 6.4–8.5 kb with the non-structural and structural proteins encoded by different ORFs. Virions are non-enveloped particles ranging from 27–40 nm in diameter with an icosahedral symmetry. In recent years, several new viruses phylogenetically distant from the 5 ICTV recognized genera as well as known norovirus genogroups and genotypes have been reported.

Aims. Update the number of calicivirus genera in the family Caliciviridae and classification of viruses in the genus Norovirus.

Methods. Calicivirus sequences in public databases from members of different genera in the family Caliciviridae and viruses from the genus Norovirus were analyzed. Greater than 60% amino acid sequence difference in the major viral capsid protein, VP1, was used to distinguish genera, genogroups and genotypes. A 2x standard deviation criterium was applied to identify new norovirus genogroups, genotypes, P-groups and P-types.

Results. Eleven calicivirus genera are now officially recognized including viruses of seven genera (Lagovirus, Norovirus, Nebovirus, Recovirus, Sapovirus, Valovirus and Vesivirus) that infect a wide range of mammals, members of two genera that infect birds (Bavovirus, Nacovirus, and members of two genera that infect fish (Minovirus, Salovirus). Within the genus Norovirus, we expanded the number of genogroups to 10 (GI-GX) and the number of genotypes to 48 (9 GI, 26 GII, 3 GIII, 2 GIV, 2 GV, 2 GVI and 1 genotype each for GVII, GVIII, GIX (formerly GII.15) and GX). Based on nucleotide diversity in the RdRp region, noroviruses can be divided into 59 established P-types (14 GI, 36 GII, 2 GIII, 1 GIV, 2 GV, 2 GVI, 1 GVII and 1 GX), 14 tentative P-types, and 2 tentative P-groups.

Discussion. This classification update of the family Caliciviridae, including noroviruses, allows improved communication among researchers of viruses that cause significant morbidity and morbidity in humans and animals worldwide.
