Anti-inflammatory Activity of the Novel Cytokine Interleukin-38 in Allergic Asthma

摘要Interleukin (IL)-38 is a newly discovered cytokine which belongs to IL-1 cytokine family. It has
been reported that IL-38 exerts anti-inflammatory activity in many inflammatory diseases. The
present study aimed to investigate the suppressive role of IL-38 in allergic asthmatic mice
model. Ovalbumin (OVA)-induced murine asthmatic model was established to evaluate the in
vivo anti-inflammatory role of IL-38, and airway hyper-responsiveness was determined by
Whole-body plethysmograph. The in vivo results showed that IL-38 injection could ameliorate
airway obstruction in asthmatic mice through inhibiting the expression of T helper (Th)2-related
cytokines IL-4, IL-5 and IL-13 in bronchalveolar lavage fluid (BALF) and lung homogenates as
well as decreasing accumulation of eosinophils in BALF (all p<0.05). Histological examination
indicated that IL-38 could alleviate lung inflammation in mice when compared with asthma
group. Also, reduced Th2 and Th17 cells and increased Treg cells were observed in mice
spleen after injection with IL-38 (all p<0.05). Taken together, our data confirmed the in vivo
anti-inflammatory role of IL-38 in murine model of allergic asthma by attenuating Th2 and Th17
cells proliferation, and promoting Treg cells differentiation.
