Molecular authentication of Ginseng constitutes in highly processed decoctions and medicinal granules

摘要Ginseng is an important herbal drug in Orient for improving vital energy (Qi) and retarding aging. Due to the morphological similarity, Ginseng species such as American Ginseng (Panax quinquefolium L.), Asian Ginseng (Panax ginseng C.A. Meyer) and Sanqi (Panax notoginseng (Burk.) F. H. Chen) are frequently substituted deliberately or unintentionally. In terms of authentication, genomic DNA provides a rich source for species identification. However, it is usually regarded as unsuitable for processed products because of DNA degradation. We set forth to investigate the availability of DNA in decoctions and medicinal granules. We have also developed multiplex polymerase chain reaction (multiplex PCR) to differentiate Asian and American ginsengs in herbal decoction. Results showed that DNA concentration increased while intactness decreased with prolonged boiling, individual herbs in decoctions as well as commercial soup products can be identified by amplifying short DNA fragments. To maximize the usefulness of currently available universal barcoding primers, we successfully employed the adaptor ligation-mediated PCR to identify Sanqi in medicinal granules. The specially designed adaptor with asymmetric strands and terminal modification avoids amplification of non-target DNA sequences. Results showed that various length of DNA fragments within the target DNA barcode regions were amplified in combination with universal primers and could be used to identify the concerned species. When this technique is combined with high-throughput sequencing technologies, the species identity and sample quality of unknown herbal products can be quickly revealed. This study thus demonstrated the presence of short DNA fragment for species identification in decoctions and medicinal granules. The work has extended the application of molecular authentication in herbal products and may be further developed into diagnostic kits for quality control and regulatory compliance.
